Description

TEM image of epon section of a freeze-substituted mouse retina illustrating the unusual distribution of heterochromatin in rod receptor nuclei where the compact heterochromatin is centrally located. Here, a 3D tomographic reconstruction (B) is shown of a boxed area (in A) containing mostly euchromatin. Technical Details

Discs were punched from freshly dissected mouse retina, cryoprotectant added, the samples cryo-fixed using high pressure freezing using a Leica EMPACT2, freeze-substituted in acetone containing OsO4, and embedded in Epon. Sections were stained with uranyl acetate and lead citrate, and observed with a Tecnai F20 TEM, and tilt series recorded with a 2k CCD camera. See also: C Kizilyaprak et al. 2010.
