Description

TEM image of epon section of a freeze-substituted mouse retina illustrating the unusual distribution of heterochromatin in rod receptor nuclei where the compact heterochromatin is centrally located. C = cytoplasm, EC = euchromatin, HCc = central heterochromatin, HCp = nuclear envelope-associated heterochromatin, NM = nuclear envelope. See Fig 1B in: C Kizilyaprak et al. 2010. Technical Details

Discs were punched from freshly dissected mouse retina, cryoprotectant added, the samples cryo-fixed using high pressure freezing using a Leica EMPACT2, freeze-substituted in acetone containing OsO4, and embedded in Epon. Sections were stained with uranyl acetate and lead citrate, and observed with a Tecnai F20 TEM, and tilt series recorded with a 2k CCD camera. See also: C Kizilyaprak et al. 2010.
