Description

Cultured HeLa cells were arrested in metaphase using nocodazole, immunostained for Bub1, Plk1 and CREST, and counter stained with DAPI. Images shown are maximum Z projections of convolved widefield images. Montage shows the effect of RNAi depletion of Bub1 and Plk1.

Technical Details

Metaphase arrested cells were formaldehyde fixed, permeabilized with Triton X-100, immunostained, and z-stack images obtained at 0.2 micron intervals with a Zeiss Axiovert 200M microscope and 63x 1.4 NA objective lens.
