Description

Image shows a z-series of a mouse ES cell carrying an alpha-globin BAC transgene array (clone 1c) hybridized with probes against the lac operator repeat (green) and the entire alpha-globin BAC sequence (red). Lac operator sequences formed foci which assumed a more interior location within the engineered chromosome subterritory formed by the BAC transgene array. DAPI counterstaining is shown in blue. Pixel sizes (xy) were 0.074 microns and z-focus steps were 0.2 microns.

Technical Details

Formaldehyde fixed cells were imaged using an IMT-2 Olympus fluorescence microscope with 60x 1.4 NA objective lens. Z-series were recorded, and the slices processed by iterative constrained deconvolution.
